Yes, it will affect your nocking point. Once you adjust poundage you must check your nock height.
Sometimes it can also cause spine issues with your arrows if you shoot on the low or high end of an arrows spine ratings.

It effects the knocking point if you don't turn the limb bolts equal amounts. Like said it's a good thing to check it when you are done. Any time you change anything on your set up you should re-check your tune. Adjusting the poundage is one of the things that can have the most profound effect on tuning.
If you are going to turn it up it may put you in a weak arrow spine condition. If you turn it down it may not hurt things as much. It is better to have a stiffer arrow than weaker one.
